摘要:The present invention provides a kind of for detecting the method for thedynamic change of the nucleic acid amount to have methylated the following steps areincluded:: 1) contact a nucleic acid sample it is doubtful with an oligonucleotide sequencebe under suitable condition that nucleic acid hybridizes comprising the nucleotide thathas methylated, the oligonucleotide sequence is characterized in that, (i) it includes:indicate first bar an of fluorophore-part, one cyclic sequence, its region with nucleotidecomplementation to an at least region for nucleic acid sample, its region is vulnerable tothe influence for making methylation, it indicates when the quencher moiety when spatialproximity is into fluorophore-part, its second bar that can quench fluorophore-part; (ii),when probe is separated from nucleic acid sample, the nucleotide for forming the firstbar can shift to the spatial proximity with the nucleotide for forming the second bar; 2)change hybridization conditions, so that oligonucleotide probe is separated frommethylate DNA, but remnants hybridize to methylate DNA; 3) change in fluorescence ismeasured.
